Without seeing I'm not 100% sure, but I am sure you will do it from inside. Might be just a couple of screws or you might have to remove an inner panel on the door. It should be pretty self evident once you look at it. Either way, it should not be very difficult.

I did not have any problem with removing the lock cylinder with the handle, but I cannot get the lock cylinder out of the plastic glove box handle.

Can it be re-keyed to match my key? How is the key pattern determined or, will several keys fit this cylinder?

Yeah, that looks like one unit from what I can tell from the front side. Any locksmith (maybe even the dealer parts guy) should be able to re-key it to your key.
