Yes. Kadonny the 2009 CTS-V (unlike the 2004-2007 CTS-Vs) does not come with run-flat tires. Contrary to CPV33's flippant reply, both the tire and the rim might be destroyed (the tire first and then the rim) if an attempt at extended driving is made on a non-run-flat tire. If you open the well on the 2009 CTS-V where the previous CTS-V had a plastic filler rather than a spare tire, you will find a small electric air pump (still no spare) with GM tire sealant that is supposed to not be a problem for the TPSs installed in the wheels.
It should also be noted that the term "Extended Mobility" was created by Mercedes and that they no longer use it for any run-flat tires that they specify. In fact, use of the term Extended Mobility has been discouraged by most car and tire manufacturers and will probably be totally abandoned pretty soon. Some tire manufacturers have two run-flat schemes (like Michelin) utilizing both stiff sidewalls (Michelin calls this ZP for zero pressure) and a plastic support installed on the rim inside the tire that supports the tire when pressure is lost (Michelin calls this the PAX system). You can use non-run-flat tires with the PAX system.
